Preliminary study of fungi from mangrove sediments as a bioremediation agent of lead (Pb) heavy metal

Abstract The mangrove ecosystem in Tapak Village is one of the coastal areas Tugu District, Semarang, Central Java and close to industrial area. This presumed be contaminated by heavy metals. study aims analysis lead metal Tapak, isolate fungi from sediments, identify potential fungus according molecular approach. sampling method used a purposive random with 4 points. isolation was carried out using dilution then cultured on general Potato Dextrose Agar (PDA) media. purified isolates were tested its ability as bioremediation agent (Pb) identified molecularly ITS1 ITS4 universal primers. results Pb content sediment samples Atomic Absorption Spectrophotometer (AAS) ranged 59.07-76.76mg/Kg, where these exceeded CCME (Canadian Council Ministers Environment) quality standards, 2001, standards 35mg/Kg. Based screening fungal media supplemented 70 ppm, total 18 purified. There 9 fastest growth observations for 7 days. One fastest-growing isolates, MT.1.E, Internal Transcribe Spacer (ITS) region homology closely related Aspergillus fumigatus which has 100% similarity. According this study, can proposed agent.
